The calibration fluid helps because you can perform a multiple point calibration, thus checking (or technically 'dialing in' your meter) to ensure a more accurate reading. Take calibrating a pH meter for example. You have 3 buffers (so that you're meter is more accurate when reading your sample. If you just calibrate with one buffer (or known solution) your reading won't be as accurate.

If you are in a pinch and can't find any RO/DI water you can use distilled water TEMPORARILY to get by. But a good RO/DI unit will be one of the best things you buy for your tank. Given that you use some decent salt and ensure the salinity is correct.
