need help with regulator

i bought a used calcium reactor with tank (just filled) and regulator. I screwed on the regulator really tight and the regulator went crazy making a really loud whistling sound. The sound is coming from a little "keyhole" on the side of the regulator. When I loosen up the knob on the regulator, the sound goes away. Am I suppose to tighten that little keyhole so air doesnt escape? Thanks guys
