Does it matter what TIME to test water
Does it matter what time I do my water test? The best time that I can do water testing is at 5-6am and 8-10pm when lights are off.

Test for what?
If pH, it will matter. My tank pH swings between 8.4 and 8.2. At night the lights are off and no photosynthetic activity is going on and the dissolved oxygen levels fall. The CO2 concentration also goes up. This causes the pH to fall to it's low of 8.2. During the day, the pH goes up due to CO2 levels falling. Alkalinity also probably varies, but probably not that much. I don't think the other parameters vary that much during the day (Nitrates NO3, Phosphates PO4). All these should be close to zero, so the avg. test kit shouldn't detect the minor changes during the day. I hope this helps.

parameter that I would test, would be calcium, alk and mag. That is all that I ever tested. The ph is always monitored by my acjr.

The time of the tests doesn't matter, per se, just the time interval since the last test.
